初步建立了复叶槭顶芽组培再生体系。N-苯基-N'-1,2,3-噻二唑-5-脲(TDZ)可诱导复叶槭顶芽分化。复叶槭顶芽的最佳增殖培养基为MS 0.1mg·L-16-BA 0.01mg·L-1NAA,最佳分化培养基为MS 0.01mg·L-1TDZ,最佳生根培养基为MS 0.1mg·L-1NAA。  相似文献

风信子试管苗,取其芽体及切块在离体培养时存在全息现象,主要表现为,在风信子芽体的不同位置的切段上。不定芽发生的数量和频率呈梯度变化;在风信子芽体的不同大小的切段上,循着切段越小不定芽的分化数越少这一规律变化,这些现象都遵循了生物全息律。适当的激素浓度对全息胚的表现具有一定的促进作用。  相似文献

姜花(Hedychium coronarium Koen.)花梗横切面整体轮廓呈椭圆形,可分为表皮、基本组织和维管束。维管束在基本组织中呈内、外两部分排列。内部维管束联结成网,形成明显外移的三束心皮背束和内方与心皮背束相间的三束隔膜束。至子房室区,心皮背束继续外移,其中主支进入花萼中脉;小分支内移,与内方一轮维管束联结,后来进入唇瓣中央及2枚侧生附属物。在花萼形成的同时,远轴面的两个隔膜中各形成一个上位腺体;同时两束远轴面隔膜束向外、两侧分别形成3束大分支,外方大分支继续外移成为2枚远轴面花瓣中脉,两侧大分支与原外方内移的子房壁维管束集合成一相连的环状维管束网,后进入唇瓣两侧;近轴面隔膜束形成3枚分支,外方分支成为近轴面花瓣中脉,两侧分支进入可育雄蕊。探讨了侧生附属物和唇瓣的来源,支持子房延长部形成的腺体为隔膜蜜腺的变异结构的观点。  相似文献

Sucrose and reducing sugar concentrations in petals of cut carnation flowers, whose life was prolonged up to 7 days by bathing stalks in sucrose solutions, were respectively 3-fold and 2-fold higher than those bathed in water. Reducing sugar concentrations were about 7-fold higher than sucrose concentrations. A study of invertase and sucrose synthase activities in flower petals of carnation and four other species of flowers revealed that both enzymes may be involved in hydrolysis of translocated sucrose. Invertase activity, while being up to 20-fold higher than sucrose synthase activity in some species was approximately comparable in others. More detailed studies on invertase from petals of 3 flower species demonstrated the presence of only the acid form of the enzyme with a Km value for sucrose of about 2.5 mM.  相似文献

The role of ethylene in vegetative bud formation was investigated using transgenic tobacco plants expressing an antisense tomato 1-aminocyclopropane-carboxylic acid synthase (ACS) gene. Northern blot hybridization showed that the accumulation of ACS mRNA was strongly reduced in the bud-forming leaf explants of the transgenic plants. Consequently, these transgenic tissues exhibited low ACS enzyme activity, 1-aminocyclopropane-carboxylic acid (ACC) content and ethylene production, and at the same time the tissue capacity to generate buds was greatly enhanced. However, it was also noted that the antisense ACS gene did not inhibit the endogenous ACS gene expression in intact transgenic tobacco plants. The growth and development of the transgenic tobacco was almost identical to control plants with respect to height, internode number, leaf morphology, and flowering time. Furthermore, mature leaves of transgenic tobacco had similar chlorophyll content, stomatal conductance, photosynthetic ability, and transpiration rates compared to control plants. These results demonstrated that ethylene plays an important role in bud formation in tobacco tissue culture.  相似文献

The effects of antibiotics commonly used in Agrobacterium-mediated transformation were studied on Pinus pinaster tissues. Embryogenic tissue growth from three embryogenic lines and adventitious bud induction from cotyledons from three open-pollinated seed families were analysed. Cefotaxizme, carbenicillin and timentin commonly used for Agrobacterium elimination, at concentrations of 200–400 mg l −1 did not inhibit the embryogenic tissue growth on filter paper nor as clumps. Adventitious bud induction and bud number were significantly reduced for one of the tested families when using 400 mg l−1 cefotaxime or timentin. The selection agent kanamycin significantly inhibited growth of embryogenic tissue on filter paper in all the embryogenic lines␣and concentrations tested (20–50 mg l−1). Kanamycin also inhibited growth of embryogenic clumps after two subcultures at 5–50 mg l−1. In␣cotyledons, kanamycin inhibited adventitious bud␣formation in the three seed families used, regardless of the concentrations tested (5–25 mg l−1). There was a significant effect of the seed family on the bud induction and the number of adventitious buds produced. From the results obtained, we propose the use of timentin to eliminate Agrobacterium in transformation experiments, at concentrations of 400 mg l−1 for embryogenic tissues and of 300 mg l−1 for cotyledons. For selection of transformed tissues carrying the kanamycin resistance gene, kanamycin should be used at 20 mg l−1 for embryogenic tissues on filter paper, at 5 mg l−1 when clumps are in direct contact with the selection medium, and bellow 5 mg l−1 for adventitious bud induction.  相似文献

Although both saproxylic longhorn beetles and hoverflies benefit from the presence of woody substrates for reproduction, they differ in their requirements for floral resources and for microbiotopes of overmature and senescent trees. This led us to expect contrasting responses between the two species groups in relation to these essential resources. We examined this prediction in 22 mature oak- and beech-dominated stands of southern Belgium by relating their species assemblages to local vegetation structure and composition, altitude and landscape composition. Stands were organised in pairs as a function of their overall dead wood supply. Free-hanging window traps, stump emergence traps and Malaise traps produced 30 longhorn beetle species (1637 individuals) and 106 hoverfly species (3020 individuals). Paired-comparisons controlling for annual variation in captures showed that, unlike saproxylic hoverflies, stands with dead wood hosted more species and individuals of longhorn beetles. Accordingly, the two species groups were found to be independent on ordination axes, responding to different sets of environmental conditions. While stands dominated by oaks with a high snag volume were highly favoured by longhorn beetles, saproxylic and threatened syrphids were limited to open-stands with large trees and a well-developed, species rich herb layer providing the floral resources required for their reproduction. Our results suggest that, when defining criteria to identify or restore important habitats for saproxylic insect conservation, variables related to different aspects of dead wood supply should not be the only criteria taken into account.  相似文献

The capacity of individual branches to store water and fix carbon can have profound effects on inflorescence size and architecture, thus on floral display, pollination, and fecundity. Mixed regression was used to investigate the relation between branch length, a proxy for plant resources, and floral display of Fouquieria splendens (ocotillo), a woody, candelabraform shrub of wide distribution in arid North America. Long branches produced three times as many flowers as short branches, regardless of overall plant size. Long branches also had more complex panicles with more cymes and cyme types than short branches; thus, branch length also influenced inflorescence architecture. Within panicles, increasing the number of cymes by one unit added about two flowers, whereas increasing the number of cyme types by one unit added about 21 flowers. Because flower production is mediated by branch length, and because most plants have branches of various lengths, the floral display of individual plants necessarily encompasses a wide range of inflorescence size and structure.  相似文献

Mammalians have a low potency for limb regeneration compared to that of amphibians. One explanation for the low potency is the deficiency of cells for regenerating amputated limbs in mammals. Amphibians can form a blastema with dedifferentiated cells, but mammals have few such cells. In this paper, we report limb formation, especially bone/cartilage formation in amputated limbs, because bone/cartilage formation is a basic step in limb pattern regeneration. After the amputation of limbs of a neonatal mouse, hypertrophy of the stump bone was observed at the amputation site, which was preceded by cell proliferation and cartilage formation. However, no new elements of bone/cartilage were formed. Thus, we grafted limb buds of mouse embryo into amputated limbs of neonatal mice. When the intact limb bud of a transgenic green fluorescent protein (GFP) mouse was grafted to the limb stump after amputation at the digit joint level, the grafted limb bud grew and differentiated into bone, cartilage and soft tissues, and it formed a segmented pattern that was constituted by bone and cartilage. The skeletal pattern was more complicated when limb buds at advanced stages were used. To examine if the grafted limb bud autonomously develops a limb or interacts with stump tissue to form a limb, the limb bud was dissociated into single cells and reaggregated before grafting. The reaggregated limb bud cells formed similar digit-like bone/cartilage structures. The reaggregated grafts also formed segmented cartilage. When the reaggregates of bone marrow mesenchymal cells were grafted into the stump, these cells formed cartilage, as do limb bud cells. Finally, to examine the potency of new bone formation in the stump tissue without exogenously supplied cells, we grafted gelatin gel containing BMP-7. BMP induced formation of several new bone elements, which was preceded by cartilage formation. The results suggest that the environmental tissues of the stump allow the formation of cartilage and bone at least partially, and that limb formation will be possible by supplying competent cells endogenously or exogenously in the future.  相似文献
